In addition, the company recently entered into a strategic partnership with Bristol Myers Squibb to develop tolerizing vaccines for up to three autoimmune diseases. On Site in Cambridge 5 days per week Role Overview We are seeking a passionate Research Associate II/Senior RA to support downstream production and process development of TCR bispecifics and MHC proteins on our Protein Engineering and Molecular Sciences team. The ideal candidate will have hands-on experience in protein purification, with preferred experience refolding proteins from inclusion bodies and/or with chemical modification of proteins. The incumbent will work at the bench to refold, purify, modify and QC TCR bispecifics and MHC proteins using biochemical methods such as ELISA, SDS-PAGE, Western, Octet and analytical methods such as HPLC. The candidate will be responsible for protein production, data analysis and presenting data to their supervisor and to the team in an effective and timely manner. Interested candidates should have strong organizational, interpersonal, verbal, and written communication skills that will enable them to integrate with our highly cross-functional team. Key Responsibilities Optimize protein refolding conditions from inclusion bodies. Develop robust processes for protein purification from mammalian expression supernatant or from refolding, using Immobilized metal affinity, ion-exchange, and size exclusion chromatography. Execute enzymatic and chemical protein modification protocols. Generate purified and modified proteins at the 5-50 mg scale and maintain inventory. Characterize recombinant proteins using SDS-PAGE, Western blot, protein quantitation, HPLC, ELISA, Octet and other biochemical/biophysical assays to determine protein purity and function. Capture primary data, establish and maintain SOPs, and document work as required in Electronic Laboratory Notebook (ELN), maintaining up to date records. Present data routinely to the Protein Sciences group and other multidisciplinary groups. Qualifications/Experience BS/MS in Biochemistry, Biology, Bioengineering, Immunology, or a similar field. 2+ years of academic or biotech/pharma laboratory experience. Experience with FPLC systems and with affinity, IEX and SEC protein purification methods is desirable. Experience with protein refolding from inclusion bodies and/or protein modification is a plus. Strong organizational and communication skills along with excellent attention to details. Enjoys working in a collaborative, fast-paced and quickly changing environment.
